摘要:本发明涉及一种可自组装成水凝胶的肿瘤杀伤性多肽,其包括C端的自组装结构域和N端的杀伤结构域,所述自组装结构域和所述杀伤结构域通过柔性结构域连接,所述自组装结构域的序列为(RADA)7‑9。所述可自组装成水凝胶的肿瘤杀伤性多肽既不影响其自组装成水凝胶的功能,又具有对肿瘤的杀伤能力。可以通过反复多次局部注射直接将缓释药剂植入到瘤体内、瘤旁或术后残腔内,不需要行开放手术暴露肿瘤,减少感染、出血等并发症,减少患者身体及经济负担。自组装多肽材料可负载化疗药物直接杀伤肿瘤,也可以负载肿瘤抗原蛋白、肿瘤裂解物或者肿瘤RNA作为肿瘤纳米疫苗使用。

Abstract: The invention relates to a tumor killing polypeptide capable of being self-assembled into a hydrogel, wherein the tumor killing polypeptide includes a C-end self-assembly structural domain and an N-end killing structural domain; the self-assembly structural domain and the killing structural domain are connected by a flexible structural domain; the self-assembly structural domain has a sequence of (RADA)7-9. The tumor killing polypeptide capable of being self-assembled into hydrogel does not affect the hydrogel self-assembling function and also has the ability to kill tumor. Through repeated multi-time local injection, slow release agents are directly implanted into tumor bodies, peritumoral parts or postoperative residual cavities, open surgery is not required to be carried out so as to prevent exposure of the tumor, infection, bleeding and other complications are reduced, and body and economic burden of patients are reduced. The self-assemble polypeptide material can be loaded with chemotherapeutic drugs for directly killing the tumor, and also can be loaded with tumor antigen proteins, tumor lysates or tumor RNA as a tumor nano vaccine for use.
