摘要:本发明涉及一种基于双酶偶联反应的高通量检测D-阿洛糖的新方法。具体涉及利用吡喃糖氧化酶和过氧化物酶偶联,检测D-阿洛糖。该方法既可用于液体培养基中反应产物D-阿洛糖的检测,也可用于检测固体培养基中反应产物D-阿洛糖的生成。当用于液体培养基的检测时,反应产物与N-(2-羟基-3-磺丙基)-3,5-二甲氧基苯胺钠(HSDA)作用产生蓝色水溶性醌类物质,利用96孔酶标板在583nm处读取吸光值。吸光值与其浓度成线性关系,能够根据标准曲线计算浓度,并利用96孔酶标板进行高通量检测。当该方法用于固体培养基中D-阿洛糖的检测时,反应产物与二氨基联苯胺(DAB)作用生成不溶于水的褐色吩嗪聚合体,利用此显色反应可在培养皿中挑出阳性菌落。 |

Abstract: The invention relates to a new method for detecting D-allose based on double-enzyme coupling high throughput, in particular to the new method for detecting the D-allose by coupling pyranose oxidase with peroxidase. The method can be used for detecting a reaction product, namely the D-allose in a liquid culture medium, and further can be used for detecting generation of the reaction product, namely the D-allose in a solid culture medium. When the method is used for detecting the liquid culture medium, the reaction product interacts with N-(2-hydroxy-3-sulfopropyl)-3, 5-dimethoxyaniline sodium (HSDA) to generate blue water-soluble quinines, and a light absorption value is read at 583nm by using a No.96 hole enzyme label plate. The light absorption value is in a linear relationship with the concentration of the liquid culture medium, and the concentration can be calculated according to a standard curve, and the high throughput is detected by using the No.96 hole enzyme label plate. When the method is used for detecting the D-allose in the solid culture medium, the reaction product interacts with diaminobenzidine (DAB) to generate a water-insoluble brown phenazine polymer, and positive bacterial colonies in a culture dish can be picked out by using a chromogenic reaction. | |
